Suwannar. 2025

Curvularia nelumbonicola Lapmak & N. Suwannar. sp. nov. (Figure 2)

MycoBank number: MB 860470

Diagnosis:—Distinguished from the closely related C. chonburiensis and C. pisi by its mostly shorter conidiophores.

Etymology:— “ nelumbonicola ” refers to the generic name of lotus which the species was isolated.

Holotype:— THAILAND. Uttaradit Province, Uttaradit Rajabhat University, Lamrang Thung Kalo Campus, isolated as an endophyte from healthy leaves of lotus ( Nelumbo nucifera), 17°36’59”N 100°08’30”E, elevation 60 m, 21 January 2022, Kodchakorn Lapmak, CMUB40077 (preserved in a metabolically inactive state), Ex-type living culture EF12A = SDBR-CMU736.

Gene sequences (from holotype):—PX125578 (ITS), PX131965 (GAPDH), and PX131967 ( TEF 1).

Endophytic from leaves of lotus ( Nelumbo nucifera). Asexual morph:—Hyphae septate, smooth, pale brown, 2.8–5.3 μm wide. Conidiophores 10–60 × 2–5 μm (x = 39.28 ± 17.5 × 3.28 ± 0.91 μm, n = 50), macronematous, mononematous, arising singly or in groups, simple or branched, septate, geniculate towards the apex, subhyaline to brown. Conidiogenous cells 3–10 × 2.5–4 μm (x = 5.86 ± 1.92 × 3.14 ± 1.02 μm, n = 50), terminal or intercalary, subcylindrical to slightly swollen, smooth-walled to slightly verruculose, pale brown to brown, with darkened scars. Conidia 10–30 × 7–15 μm (x = 21.58 ± 5.14 × 11.81 ± 2.00 μm, n = 50), brown to dark brown, smooth-walled, mostly slightly curved, sometimes ellipsoidal to barrel-shaped, 2–3-distoseptate, tapering toward rounded ends, third cell from base often larger, apical and basal cells paler than the middle cells; hilum non-protuberant, flat, darked and thickened, 1.3–4 μm wide. Chlamydospores and microconidia were absent.

Sexual morph:—Unknown.

Culture characteristics:—Colonies diameters after one week at 25 °C on PDA, OA, and PCA were 76–79, 75–78, and 72–75 mm, respectively. Colonies on PDA were greyish yellow, with sparse white aerial mycelia, velvety, circular, flat, margin slightly irregular and fimbriate; reverse olive grey. Colonies on OA were greyish yellow, with sparse white aerial mycelia, velvety, circular, flat, margin slightly irregular and fimbriate, olivaceous gray at center; reverse olive grey. Colonies on PCA were olivaceous gray to greenish grey with sparse gray aerial mycelia, circular, flat, margin slightly irregular and fimbriate; reverse greenish grey. Sporulation was observed on all three media after one week.

Cardinal temperature for growth:—Optimum 25 °C, maximum 37 °C, minimum 10 °C.

Distribution:—Known only from Thailand.

Additional culture examined:— THAILAND. Uttaradit Province, Uttaradit Rajabhat University, Lamrang Thung Kalo Campus, isolated as an endophyte from leaves of lotus ( Nelumbo nucifera), 17°36’62”N 100°08’35”E, elevation 62 m, 15 October 2022, Kodchakorn Lapmak, living culture EF60B = SDBR-CMU737, GenBank number PX126166 (ITS), PX131966 (GAPDH), PX131968 ( TEF 1).

Note:—Phylogenetically, C. nelumbonicola forms a monophyletic clade that is sister to type strains of C. chonburiensis (MFUCC 16-0375), C. hominis (UTHSC 09-464), C. muehlenbeckiae (CBS 144.63), C. platzii (BRIP 27703b), C. pisi (CBS 190.48), and C. plantarum (CGMCC 3.19342), and C. lycopersici strain 11 with 79% BS and 0.96 PP support values. (Figure 1). Morphologically, the 2–3-distoseptate conidia of C. nelumbonicola differ from those of C. hominis (4–5-distoseptate), C. muehlenbeckiae (4-distoseptate), and C. plantarum (4-distoseptate), and C. platzii (9–13-distoseptate) (Madrid et al. 2014, Tan et al. 2018, Raza et al. 2019) (Table 3). Curvularia nelumbonicola resembles C. chonburiensis and C. pisi in having 2–3-distoseptate conidia. However, the shorter conidiophores in C. nelumbonicola (10–60 μm long) distinguish it from C. chonburiensis (70–200 μm long) (Tibpromma et al. 2018) and C. pisi (50–210 μm long) (Marin-Felix et al. 2017). There is currently limited information on the morphological description of C. lycopersici . A pairwise nucleotide comparison of GAPDH sequences shows that C. nelumbonicola differs from C. chonburiensis, C. hominis, C. muehlenbeckiae, C. platzii, C. pisi, and C. plantarum by 2.12%, 3.59%, 3.92%, 5.39%, 4.08%, and 3.10%, respectively (Table 4).

https://treatment.plazi.org/id/BB0287D5CE20FFC7FF553CDBFCB522F3	Public Domain	No known copyright restrictions apply.
