DESCRIPTION OF NOCARDIOIDES VASTITATIS SP. NOV.
Nocardioides vastitatis (vas.ti.ta′ tis. L. masc. gen. n. vastitatis of a desert).
Cells are Gram-stain-positive, aerobic, non-motile, nonspore-forming and rod-shaped (approximately 0.4– 0.7×1.1–2.3 µm). Colonies on ISP 2 agar are circular, smooth, opaque, creamy white in colour. Grows well on ISP 2, TSA, R2A agar and nutrient agar. Poor growth occurs on ISP 3, ISP 4, ISP 5 and ISP 7 agar. Growth occurs at 10– 37 Ǫ C (optimum, 28–30 Ǫ C), pH 6.0--9.0 (optimum, pH 7.0) and with NaCl concentrations of 0–3 % (w/v; optimum, 0 %). Catalase-positive and oxidase-negative.
Hydrolyses Tween 20 and Tween 40, but does not hydrolyse casein, gelatin, Tween 80, carboxymethyl-cellulose, skimmed milk, starch and aesculin. Negative for nitrate reduction, indole production, glucose fermentation, H 2 S production, urease and arginine dihydrolase. Assimilates D-mannitol and malate. Does not assimilate D- glucose, L- arabinose, D- mannose, N - acetyl-D- glucosamine, maltose, potassium gluconate, caprate, adipate, citrate and phenylacetate. Positive for alkaline phosphatase, esterase (C4), esterase lipase (C8), leucine arylamidase, acid phosphatase, naphthol-AS-BI-phosphohydrolase, b- galactosidase, a- glucosidase and b- glucosidase, but negative for lipase (C14), valine arylamidase, cysteine arylamidase, trypsin, a- chymotrypsin, a- galactosidase, b- glucuronidase, N -acetyl-b- glucosaminidase, a- mannosidase and a- fucosidase. Acids are produced from L- arabinose, D- xylose (weakly), D- glucose, D- fructose, L- rhamnose, inositol, mannitol, aesculin, sucrose (weakly), inulin (weakly) and raffinose (weakly). The cell-wall peptidoglycan contains LL- diaminopimelic acid as the diagnostic diamino acid. The predominant menaquinone is MK-8(H 4). The polar lipids comprise DPG, PG, PI, an unidentified phospholipid and three unidentified lipids. The major whole-cell fatty acids (>10 %) are iso-C 16: 0 and 10-methyl C 18: 0 (TBSA). The G+C content of the genomic DNA is 70.4 mol%.
The type strain, 21Sc5-5 T (=JCM 33365 T =CGMCC 4.76 08 T), was isolated from soil sampled at the rhizosphere of Inula salsoloides collected in the Taklamakan desert in Xinjiang Uygur Autonomous Region, PR China .
